EDIT: i was able to do it for grid issue by going into the /mui config and unchecking auto switch profile for grid but when i do it for unit frames it still dosnt work, maybe because i am using the OLD unitframes plugin ?
It depends how old the unit frames plugin is but I really think that should not matter if the UI is updated to...

When I disable that add-on, it tells me another one is doing same, etc. Also, how can I make it so that I can right click a buff in combat to remove the buff (i.e. symbiosis)...

That is because you have the bottom of the UI Expanded I bet. If that is the case then the UI will think Bar 9 is suppose to be showing. You can alter which 2 bars you want showing when the UI is expanded by typing "/mui config" and scroll...
